肌氨酸在人體尿液中被發現可能為惡性前列腺癌的重要指標分子,人體中的新喋呤濃度增加會造成應激活化及氧化劑之間平衡的重要,進而使癌症腫瘤增生的另一關鍵。在本研究中希冀以聚乙烯乙烯醇高分子拓印目標分子肌氨酸、新喋呤與肌酸酐,並使用溶劑蒸發法讓聚乙烯乙烯醇高分子在基材上形成薄膜,使得目標分子固定在薄膜上,移除目標分子使得拓印後高分子能對特定的目標分子具有辨識效果,其拓印後的肌氨酸與新喋呤薄膜,分別對於乙烯莫爾比例32及27的聚乙烯乙烯醇有較好的拓印效率,拓印效率可達2.08與2.05。隨著聚乙烯乙烯醇生物相容性高和易成膜的特性,使得此種薄膜適合做為生物感測器上的受體材料,並可製作成工作電極進行非侵入式的尿液檢測,結合自製的電化學感測器,對肌氨酸濃度進行非侵入性檢測,偵測極限可達0.01 ng/mL,其sensitivity可達1.5uA/cm2,在進行additional recovery的可轉換出尿液檢體中肌氨酸濃度。為了使量測方法的靈敏度更高與更微小化的感測裝置,將藉助強制對流的方式,使電化學反應能順利的在電極表面產生,利用電極微小化的方式將電極固定在玻璃基板上作為電化學感測器電極整合微流體通道。 Sarcosine was identified as a differential metabolite that was highly increased during prostate cancer progression to metastasis and can be detected non-invasively in urine. The higher concentration of neopterin production is associated with increased production of reactive oxygen species, and might be the best predictors of adverse outcome in patients with various types of cancer. In this work, the sarcosine, neopterin and creatinine is used as the target molecules and then imprinted into poly(ethylene- co-vinyl alcohol) via solvent evaporation. Target molecules on membrane are then removal, make sure those membrane can recognize the unique molecules. The preliminary binding measurements of sarcosine and neopterin molecules to the sarcosine-, neopterin- and non-imprinted EVAL polymers were performed the highest imprinting effectiveness for sarcosine and neopterin was obtained with 32mol%and 27 mole % of ethylene, respectively. The highest imprinting effectiveness for each target molecule was 2.08 and 2.05, respectively. With the high biocompatibility and easy manufacture of EVAL thin films, these membranes should be very suitable for use as sensing materials in biosensors. Those molecule-imprinted electrode were prepared for the detection of sarcosine, neopterin and creatinine in real sample (urine). The homemade potentiostat can be integrated with the molecularly imprinted polymer sensing system for actual urine sample analysis. Moreover, the limit of detection (LOD) of this electrode for sarcosine is less than 0.01 ng/mL, the sensitivity up to 1.5 ?A/cm2.In order to better measurement methods and more sensitive sensor miniaturization of devices (30 × 1 mm), will be the way with forced convection, so that make sure electrochemical reaction at the electrode surface generates. Uses miniaturized electrodes fixed to the glass substrate as the electrochemical sensor electrode, combined with PDMS polymer microfluidic channel fabricated.
