本菌株是由台灣南部土壤,由鹼性培養基篩選而來,經新竹食工所菌種中心鑑定,確定為放線菌,為革蘭氏陽性菌。菌株經有機溶劑萃取後,其粗萃液於分光光度計下作全光譜掃描,在480 nm有最大峰值。最適萃取液為氯仿:甲醇 = 1:1;最適培養基為0.8 % Glucose、1.8 % Yeast extract、2.0 % NaCl;培養溫度為30℃;培養時間為72小時;轉速150 rpm。將粗萃液通入Silica gel 60管柱,收集紅色部分,紅色部分以減壓濃縮抽乾,加入二氯甲烷與甲醇 = 1 : 2,靜置於4℃,48小時後結晶析出。薄層色譜中,其Rf值為0.3,且UV254和UV366下亦無其它點存在。結晶物全光譜掃描在480 nm有最大峰值。紅色色素的抗氧化能力IC50值為每毫升15微克(Vitamin C IC50值為每毫升52微克、Vitamin E IC50值為每毫升7微克);紅色色素具光穩定性及熱穩定性,分別用40w日光燈管照射2小時與60℃處理後,色素仍處於穩定狀態。 The strain of salt-tolerant actinomycete NO.38 strain was isolated from outdoor soil of southern Taiwan. Bioresource Collection and Research Center (Hsinchu, Taiwan) was identified the strain as actinomycete. The crude extract by organic solvent (Chloroform : Methanol = 1:1) extracted revealed a maximum peak on 480 nm of wavelength scanning on spectrophotometer. The composition of optimal medium for the strain to produce the red pigment contained 8 g of glucose, 18 g of yeast extract and 20 g of sodium chloride in 1 liter of distilled water. The culture was incubated at 30℃ on rotary shaker at 150 rpm for 72 hours. Base on polarity of chemical solutions, we used silica gel column chromatography to exclude most impurities and collected the red fraction. The red fraction dried by vacuum desiccator. The red dried fraction was added solution (Dichloromethane Methanol = 1:2), and placed it in a refrigerator for crystallization. The crystalline was washed with methanol and redissolved with chloroform. After thin layer chromatography, the Rf (Retardation factor) value of the red pigment is 0.3, and there had not found any other bands under UV254 and UV366. The IC50 antioxidant activity of the red pigment is 15
