    摘要註: 本研究希望能了解在熱壓力的作用下硫辛酸對不同骨骼肌群HSP70及GLUT4的影響。在本論文中SD大鼠以腹腔注射的方式施打硫辛酸(50 mg/kg b.w.),連續施打三周。為了評估硫辛酸的保護效果,我們將SD大鼠置入40℃乾式循環烘箱進行熱處理15分鐘,三周後施以過量麻醉劑犧牲大鼠並將伸趾長肌(extensor digitorum longus)、橫膈肌(diaphragm)、腓腸肌(gastrocnemius)、比目魚肌(soleus)取出並以西方墨點法進行熱休克蛋白70(heat shock protein, HSP)、葡萄糖轉移子4(glucose transporter 4, GLUT4)的蛋白質分析。結果發現,不論是給予硫辛酸或熱休克(heat shock)在SD大鼠四種骨骼肌的HSP70表現都會上升,特別在大鼠比目魚肌表現明顯增加。另一方面,在大鼠比目魚肌施予熱休克處理可提升GLUT4表現,而且在大鼠四種骨骼肌中硫辛酸都無法誘導GLUT4表現提升。   本研究另外將C2C12小鼠肌纖維母細胞株分別培養於5mM和25mM葡萄糖濃度的培養環境中來探討GLUT4和HSP70表現情形。利用西方墨點法結果發現,C2C12 myotubes培養於不同葡萄萄濃度的GLUT4表現則無差異,而在高醣培養環境下投與硫辛酸作用3小時HSP70表現高於低醣培養環境,特別在熱休克下硫與硫辛酸後HSP70提升更為明顯。在細胞修復(wound healing)結果發現,硫辛酸在25mM葡萄糖濃度下作用3小時可誘導C2C12受傷害後的修復能力。綜合以上結果顯示,在骨骼肌上不論是in vivo或in vitro的實驗硫辛酸皆可誘導HSP70表現提升,另外在大鼠比目魚肌上熱休克誘導GLUT4則可在未來研究中進一步討論。 The aim of this study is to understand the expressive effects of lipoic acid in different skeletal muscle groups of HSP70 and GLUT4 under heat shock. In this study, 50mg/kg lipoic acid was applied to SD rats by intraperitoneal injection for three weeks. To evaluate the protect effect of lipoic acid, SD rats were further treated at 40℃ in circulated drying oven for 15 mins. After 3 weeks, SD rats were sacrificed and gastrocnemius, soleus, diaphragm, extensor digitorum longus were obtained. HSP 70 and GLUT4 proteins were assayed by western blotting. Our data showed both lipoic acid and heat shock up-expressed HSP70 in four skeletal muscles, especially highly expressed in soleus. On the other hand, GLUT4 can up-expressed in soleus by heat shock treatment. Lipoic acid could not evoke the expression of GLUT4 in all skeletal muscles studied today. Besides, the expression of GLUT4 and HSP70 were also studied in C2C12 myotubes culture with containing 5 mM and 25 mM glucose condition. The results of western blots show HSP70 has higher expressed in 25 mM glucose condition and there is no difference in the expression of GLUT4 with 5 mM or 25 mM glucose condition. Moreover, lipoic acid induced the expression of HSP70 only in 25 mM glucose condition for 3 hrs, escpecially highly expressed in heat shock treatment . In wound-healing study, it is also showing lipoic acid induced wound closure in 25 mM glucose condition for 3 hrs. Collectively, our results indicate lipoic acid could up-expressed HSP70 in vivo or in vitro in skeletal muscles. The result of expressed GLUT4 only in soleus by heat shock treatment could be good information for further study.
