    摘要註: 我們實驗室已成功利用電噴灑游離源之霧化氣體開發出文氏管電噴灑游離質譜法 (Venturi electrospray ionization mass spectrometry, VESI/MS),此項質譜法具有快速、簡單、樣品使用量少與可進行線上即時偵測之能力等優點。為了瞭解VESI/MS未來可能之應用性,本研究以單重和雙重進樣方式之VESI/MS偵測一系列性質不同之實驗,凸顯線上即時偵測於反應動力學研究上之優點。以五種溶劑測試結果顯示,VESI裝置內溶液之平均流速約為9~32 μL/min,10~20秒即可將分析液傳送至游離源進行分析,具有線上即時偵測之功能。使用單重進樣方式之VESI/MS (sVESI/MS) 探討苯並咪唑化合物之合成反應,分析不同溶劑或有無酸催化下,單取代與雙取代基產物之消長變化,可以用來判斷何者是動力學有利或熱力學有利之產物。探討藍瓶反應之應用,則是依據還原態亞甲基藍之選擇離子層析(EIC)圖,展現還原劑濃度與氧氣對此氧化還原反應速率之影響。有別於探討化學反應機制,sVESI/MS轉而研究茶葉於不同沖泡條件下所溶出物質之變化,EIC圖清楚顯示發酵程度、沖泡溫度與沖泡次數對茶葉溶出物質之影響。利用sVESI/MS具有自動進樣和分析液只需10 μL之優點,我們以sVESI/MS進行水果表皮農藥殘留之分析,證實洗滌水果雖有清潔作用,但無法完全移除農藥之殘留。苯並咪唑化合物之合成反應改以雙重進樣方式之VESI/MS (dVESI/MS) 偵測時,EIC圖證實反應開始數秒之內中間產物已生成,再隔數秒後產物也開始生成。藉由dVESI/MS 可以使用post-reactor方式添加氨水或醋酸於分析液中之優勢,我們成功提升偵測茶湯內容物時,茶氨酸、咖啡因和兒茶素之訊號強度。 Extended utilization of nebulizer gas of electrospray ionization source, we had developed Venturi electrospray ionization mass spectrometry (VESI/MS). The advantages of this method are rapid, simple, small sample amount, and on-line real-time monitoring. To explore potential applications of this method in the future, we examined a variety of applications of VESI/MS. Sample inlet methods of VESI/MS can be single-inlet or dual-inlet. Information provided by VESI/MS was especially favored for studies of reaction kinetics.Measurements of five different solvents showed that the flow rate for delivering solution was ranged from 9 to 32 μL/min. Delivery time of solution was 10~20 seconds. Hence, VESI/MS was suitable for on-line real-time analysis. Single-inlet VESI/MS (sVESI/MS) was applied on monitoring synthesis of benzimidazole. Productions of monosubstituted and disubstituted final products upon different experimental conditions were recorded continuously. Experimental conditions that studied were solvent effect and acid catalysis. Results were conducted to justify which product was kinetic-controlled product. sVESI/MS was also applied on study of blue bottle reaction. Fluctuation of reduction-state methylene blue upon periodic redox reaction was clearly demonstrated by extracted ion chromatogram (EIC). The function of oxygen and effect of fructose concentration on the reduction rate were also shown.Besides reaction mechanism, sVESI/MS can be used to investigate compounds released from tea leaves upon different drewing conditions. Effects of drewing temperature and number of drewing and extend of fermentation were examined. sVESI/MS was an automatic sampler and required small sample amount (10 μL). It was suitable for preliminary inspection of pesticide residue of fruits. We examined the washing procedures for cleaning out pesticide residue of fruits. Results indicated that the pesticide residue can be removed to an extent, but not all. Synthesis of benzimidazole was also studied by dual-inlet VESI/MS (dVESI/MS). EICs obtained from dVESI/MS showed that reaction intermediate was formed in few tens of seconds after reaction initiated. Final products were formed in seconds after the formation of intermediate. dVESI/MS had a capability to perform post-reactor technique. By on-line adding ammonia or acetic acid into solution that brewing from tea, the signal intensity of theanine, caffeine, and catechin in mass spectrum were improved with dVESI/MS.
