生長素釋質(ghrelin)主要是由胃上皮黏膜細胞所分泌的一種內源型生長荷爾蒙分泌素(growth hormone secretagogue, GHS)。本研究的目的是評估豬ghrelin對於發炎細胞激素之調節及做為新穎佐劑之可行性。分析豬不同組織細胞ghrelin基因的表現情形,結果顯示在胃組織表現量遠高於其他組織。將preproghrelin、proghrelin及ghrelin基因選殖入E. coli表達系統,以IPTG誘導蛋白表達後進行純化,利用西方點墨法確認融合蛋白能正確表現。以脂多醣誘導產生發炎相關細胞激素之試驗,顯示proghrelin具有抑制TNFα及IL-1β表現的能力,推測能降低發炎反應產生。融合蛋白刺激巨噬細胞試驗,皆能顯著增加細胞激素IL-12的表現,推測能誘發Th1免疫途徑。以ovalbumin為抗原搭配融合蛋白做為佐劑,經由肌肉注射免疫小鼠,抗體效價可達到51200以上。抗體亞型分析顯示proghrelin能提升IgG2a的表現量,推測在動物體內能誘發Th1免疫途徑。免疫後小鼠進行脾臟細胞增生試驗與細胞激素表現量分析,結果顯示proghrelin、ghrelin及syn-ghr能夠有效同時誘發Th1和Th2免疫反應,綜合上述結果,proghrelin最具有開發為細胞性免疫佐劑之潛力。 Ghrelin is a kind of endogenous growth hormone secretagogue (GHS) which is secreted by stomach mucosal epithelial cells. The purpose of this study is to evaluate the modulation of inflammatory cytokines by ghrelin and the possibility of ghrelin being a novel adjuvant. Analysis of the expression of ghrelin in various swine tissue cells, it showed that the expression in stomach tissue was significantly higher than in other tissues. The gene of preproghrelin, proghrelin, and ghrelin was cloned into E. coli expression system, induced protein expression by IPTG and purified. The fusion protein expression was identified by Western blot analysis. The experiment of lipopolysaccharide-induced inflammatory cytokines expression showed that proghrelin could inhibit the expression of TNFα and IL-1β. It indicates that proghrelin could reduce the inflammation. The expression level of IL-12 was significantly enhanced by these recombinant proteins stimulated macrophage. It indicates that the immune response towards to Th1 pathway. Using ovalbumin as antigen combined with these recombinant proteins to evaluate their adjuvanticity in mice through intramuscular injection. The antibody titers specific to ovalbumin could reach higher than 51200. The results of IgG subclass analysis showed that proghrelin could enhance the expression of IgG2a. It indicates that could induce Th1 pathway in vivo. The immunized mice were used for the splenocytes proliferation assay and cytokines analysis. The results showed that proghrelin, ghrelin, syn-ghr could effectively induce both of Th1 and Th2 immune responses. To sum up, proghrelin has the potent potential for the development of adjuvant for the induction of cellular immunity.
