一些植物種子種皮的最外層含有黏質,此黏質具有吸取水分、黏附土壤或動物及確保種子萌芽等多項生理功能。由於白柚 (Citrus grandis Osbeck)種子浸潤水後表層呈現透明的黏質物,而且經ruthenium red染色後呈紅紫色,確認該黏質物的主要成份是酸性多醣。本論文期望藉由組成與特性分析,評估柚籽表面的酸性多醣是否具有應用於食品及化妝品的潛力。以室溫的水萃取柚籽表面黏質物 (water extract,WE),產率約3%。WE具有高黏度特性,1% (w/v)水溶液可達6000 cP以上。WE經由陰離子交換層析 (DE-52)分離為主要的兩個fraction,分別為於二次水流洗出的Fraction A,以及於0.10 The outer layer of the seed coat in some plant seeds contains large quantities of mucilage. Mucilage in these seeds is thought to perform a variety of physiological functions, including the absorption of water, attachment to soil substrates and animal vectors, protection the seed and its germination. Following the imbibitions of water, the outer layer of shaddock seed (Citrus grandis Osbeck) produces a transparent mucilaginous gel, which appears purplish red after stained with ruthenium red, identifying the gel as an acid polysaccharide. The purpose of this thesis was to analysis the components and properties of shaddock acid polysaccharide, and estimate its potential applicability in food and cosmetics. Mucilage was extracted using deionized water at room temperature (water extract, WE), yielding approximately 3% (w/w). WE displayed high viscosity of 1% (w/v) exceeding 6000 cP. WE was separated into two major fractions by anionic exchange chromatography on DE52-cellulose, namely, Fraction A and Fraction B, collected during elution with water and 0.10–0.18 M NaCl gradient, respectively. Fraction B was the major component of WE, with a high content of uronic acid, and the molecular weight ranging from 73 to 242 kDa. The major monosaccharides identified in Fraction B were rhamnose, arabinose, galactose, glucose, mannose or xylose, galacturonic acid and glucuronic acid. Neither WE nor Fraction B displayed significant scavenging effects on DPPH radicals, and the Fe+2 chelating abilities were considerably weaker than that of EDTA. Nonetheless, both WE and Fraction B demonstrated high superoxide dismutase activity, and high hydration capacity of 9.4 and 15.2 mL/g dry weight, respectively. Neither WE nor Fraction B demonstrated cytotoxic effects on NIH-3T3 cells.
