在近十幾年中,疾病診斷偵測方法的改進一直著重在增加靈敏度、提昇偵測極限、以及縮短診斷時間,而我的研究目的在於放大來自於抗體抗原錯合物的訊號,所使用的訊號放大介質為卵白素修飾微球體以及生物素修飾微球體。其策略假說為如果目標物被抗體修飾表面捕捉,將會觸發微球體間的聚合反應進而放大訊號達到視覺偵測。聚合反應的觸發是來自於卵白素-生物素間的反應,此反應為俗知的強非共價鍵結之一;而視覺偵測的達成主要是藉由自組裝的微球體,形成肉眼或是顯微鏡可觀察到的現像。此種簡易的偵測方式將可能在少許的操作步驟下,就能運用在辨識微生菌的感染。 In the recent decades, the improvement of the diagnostic detection method is focusing on raising the sensitivity, enhancing the detection limit, and speeding up the diagnosis process. The purpose of my research is to amplify the initial signals from the antibody-antigen complexes. The signal enhanced media are the streptavidin modified microspheres and the biotin coated microspheres. The hypothesis of our strategy is if the analytes are detected by the antibody coated surface, polymerization of the microspheres is triggered and can amplify the initial signals and lead to visual detection. The polymerization is achieved by the streptavidin-biotin interaction, which is known to be one of the strongest non-covalent bonds in nature. The visual detection of the targets is accomplished by the self-assembly of functionalized microspheres which can be observed by naked eye or a commercial microscope. This simple detection method may be used to identify infectious microorganisms in few steps.
