Summary:
一系列於本實驗室合成的小分子藉由高效液相層析與晶體結構分析以探討其詳細的掌性分離機制。透過特殊設計後的結構,我們排除其他潛在足以影響π-π交互作用的因素,並將重點放置於苯環上取代基所造成的影響。與本實驗室之前探討同分子間結晶性質的研究相比,本研究著重於不同取代基分子間相對的π-π交互作用。藉由高效能液相層析,我們得以探討不同取代基的分析物與結構相似的市售管柱間固定相的交互作用力。同時,我們設計了一系列的共結晶以探討分析物與管柱固定相間的相對構型以及交互作用力強度。最後,結合高效能液相層析與共結晶所得到的結果,我們發現了一些影響取代基效應的關鍵因素,而這些因素在掌性固定相的設計上將會有決定性的影響。 A series of small-molecules synthesized in our lab had been discussed using HPLC-UV and crystallographic analysis in order to investigate the detailed chiral separation mechanism. With the designed model, we excluded other factors that are potentially influential to the strength of π-interaction and focus on the substituent effect. Compared to our previous work on probing of the crystallographic properties of dimers, this work primarily focuses on the relative π-π interactions between molecules with different substituents. With the help of HPLC-UV analysis, we evaluated the interactions among various substituted compounds on the structure alike commercially available column. Also, we have designed a series of co-crystal to assess the relative geometry and interaction strength between analytes and chiral stationary phase (CSP). Combine the co-crystal results obtained so far with HPLC-UV data, we have found some crucial factors related to substituent effect, and may have a decisive influence on CSP design.
