Carbon dots (CDs) are used for biosensing because of their practical application, fluorophoric nature, low toxicity, and abundant precursors. The fluorescence of CDs was used for drug detection when combined with a DNA aptamer that is specifically coded for a target molecule. The negatively charged cocaine DNA aptamer electrostatically binds to the positively charged CD made from malic acid and polyethyleneimine. The DNA aptamer effectively quenched the CDs fluorescence. Once the cocaine was added to the CD-aptamer complex, the aptamer released from the CD as the aptamers preferentially bind to the cocaine molecule. The CD fluorescence was recovered indicating the presence of cocaine. The CD-aptamer complex was tested for specificity by using lidocaine and procaine which are similar in structure to cocaine. Results show that the aptamer has a higher affinity for the drug molecule by uncapping itself from the CD. The uncapping of the aptamer results in an increase in fluorescence intensity. The increase isn't currently linearly proportional to the amount of drug dosed to the system. The salt concentration of the aptamer solution could possibly be interfering with the linearity which is going to be the focus of future research.
