Polysaccharides are complex biomacromolecules known to provide a wide range of biological functions such as cellular structural support, energy storage, and cellular communication. In recent years, several polysaccharides were found to play a crucial role in biological activities such as antioxidant, anticoagulant, prebiotic, and immunobiological effects. The biological activities of such polysaccharides are heavily correlated with their structural compositions. However, most studies have focused largely on the biological effects with little to no description of the structural characteristics of the bioactive polysaccharide. This is attributed to the inherent structural variability in the monosaccharide composition and arrangement, glycosidic linkage types, and degree of branching of polysaccharides. In this dissertation, novel platforms for characterization and identification of polysaccharides will be introduced. The presented methods are a significant advancement over conventional methods from the 1960s. In Chapter 1, information on the structure and function of polysaccharides and current strategies for their characterization will be discussed. Chapter 2 presents a novel method for the dissociation of polysaccharides into oligosaccharides and their identification using mass spectrometry. Chapter 3 discusses the application of the technique discussed in Chapter 2 towards rapid identification of polysaccharides using a single instrument platform. Chapter 4 leverages the technique from Chapter 2 in subjecting polysaccharides to alternative metal catalysts to study metal specificity during oxidative dissociation. Chapter 5 presents the first extensive synthetic glycosidic linkage library for the comprehensive analysis of complex polysaccharides.
